Isolation, production, purification, and estimation of Vitamin B12 from Streptomyces organisms with antibiotic productivity from the soil of Anantapur region

Abstract


Streptomyces is the genus, family of Streptomyceae belonging to the order Actinomycetales and the
class Schizomycetes, having highest number of bio-molecule productivity among the organisms isolated till
date. Ascertaining the importance of microbial production and unexplored microbial potential of Anantapur,
Andhra Pradesh, region made us to search for the Streptomyces species of this region with dual productivity of
Vitamin B12 with antibiotic productivity. With this aim the present work was carried out with the initial screening
of the soil for antibiotic producing Streptomyces by crowded plate method using M2 agar media which reported as
a selective media for Streptomyces. Production was carried out using the same media and Vitamin B12 productivity
was estimated using High-performance liquid chromatography (HPLC) method after purification from the
fermented broth. Out of our search, we successfully isolated 10 Streptomyces species with the capability for
dual production of Vitamin B12 and antibiotics. Among these, strains 1A10, 1B8, 2B5, 1B5, and 4B8 exhibited
the highest Vitamin B12 productivity based on HPLC estimation. Concerning antibiotic production, strains 1B8,
2B5, 1B5, and 4B8 showed significant zones of inhibition against the test organisms. Notably, strains 1B5 and
4B8 demonstrated the highest dual productivity, combining substantial Vitamin B12 production with pronounced
antimicrobial activity. Strain 1B5 stood out with the highest Vitamin B12 concentration and broad-spectrum
antimicrobial effects, while strain 4B8 also exhibited significant dual productivity. These findings underscore 1B5
and 4B8 as exceptional candidates for further development due to their superior dual productivity in both Vitamin
B12 synthesis and antimicrobial efficacy.
